Letrozole is a prescription medication classified as an aromatase inhibitor (AI). It is most commonly used in the treatment of hormone receptor–positive breast cancer in postmenopausal women. A 2.5 mg tablet is the standard pharmaceutical strength for medical use.
Letrozole works by inhibiting the aromatase enzyme, which is responsible for converting androgens (such as testosterone) into estrogen. By reducing estrogen production, letrozole lowers circulating estrogen levels in the body.
Drug Class: Non-steroidal aromatase inhibitor
Primary Action: Blocks conversion of androgens to estrogen
Result: Significant reduction in systemic estrogen levels
Because estrogen can stimulate certain breast cancer cells to grow, reducing estrogen levels can help slow or stop tumor progression in hormone-sensitive cases.
Letrozole is commonly prescribed for:
Treatment of hormone receptor–positive breast cancer in postmenopausal women
Extended adjuvant therapy after tamoxifen
Ovulation induction in certain fertility treatments (off-label use under medical supervision)
By lowering estrogen levels, letrozole may cause:
Joint pain or stiffness
Hot flashes
Fatigue
Bone density reduction (increased osteoporosis risk)
Elevated cholesterol in some individuals
Headaches
Because estrogen plays an important role in bone health and cardiovascular protection, long-term use requires medical monitoring
