🧬 RETATRUTIDE — THE TRIPLE-AGONIST BEING STUDIED FOR WEIGHT MANAGEMENT
Retatrutide is an investigational medication being developed by Eli Lilly for obesity and other metabolic conditions. What makes retatrutide different from many existing GLP-1 medications is that it targets three different hormone receptors at the same time: GLP-1, GIP, and glucagon receptors.
These pathways are involved in appetite, blood-sugar regulation, energy balance, and metabolism. Researchers are studying whether activating all three receptors can produce greater metabolic and weight-management effects than medications targeting only one or two pathways.
🔥 WHAT IS RETATRUTIDE BEING STUDIED FOR?
Clinical research has primarily investigated retatrutide for obesity and weight management, including its effects on body weight, waist circumference, metabolic health, and related conditions.
Results have been significant. In Lilly’s Phase 3 TRIUMPH-1 trial, adults with obesity or overweight who received the 12-mg target dose achieved an average 28.3% reduction in body weight at 80 weeks. Individual results varied, and clinical-trial results shouldn’t be interpreted as a guarantee of weight loss for any individual.
💉 HOW WAS RETATRUTIDE DOSED IN CLINICAL TRIALS?
Retatrutide has been administered as a subcutaneous injection once weekly in clinical studies. Importantly, researchers generally didn’t start participants immediately at the highest target dose.
In the Phase 3 program, participants began at 2 mg once weekly, and doses were gradually increased at four-week intervals toward assigned target doses such as 4 mg, 9 mg or 12 mg once weekly.
That gradual escalation is important because gastrointestinal side effects tend to occur more frequently while doses are being increased.
🤢 POSSIBLE SIDE EFFECTS
Side effects reported during clinical research have included nausea, diarrhea, vomiting, constipation and decreased appetite. Because retatrutide activates the glucagon receptor in addition to GLP-1 and GIP receptors, researchers are also continuing to evaluate its broader metabolic and cardiovascular effects.
