Retatrutide is an investigational (not yet FDA-approved) medication being studied for obesity and type 2 diabetes. It’s currently in clinical trials and not commercially available through regular pharmacies.
Retatrutide activates three receptors:
GLP-1 → reduces appetite, slows digestion
GIP → improves insulin response and metabolic efficiency
Glucagon → increases energy expenditure (burns more calories)
That third mechanism (glucagon activation) is what separates it from:
Semaglutide
Tirzepatide
Retatrutide potentially increases both appetite control and metabolic rate.
In clinical trials, dosing is gradually increased to reduce side effects.
5 mg would generally be considered a mid-range titration dose, depending on the protocol being followed.
Because it is still experimental:
There is no standardized public dosing protocol
Long-term safety data is still being collected
Products outside clinical trials are unregulated
In Phase 2 studies:
Up to 24%+ body weight reduction
Significant fat mass loss
Improved blood sugar control
Noticeable waist circumference reduction
These results were among the strongest seen in weight-loss medications to date.
Nausea
Vomiting
Diarrhea
Strong appetite suppression
Elevated heart rate (due to glucagon activity)
Side effects tend to increase as dosage increases.
